产品描述
壬基吖啶橙(Nonyl Acridine Orange,NAO),CAS NO. 75168-11-5,是一种吖啶橙(Acridine Orange,AO)衍生物,通常用作完整细胞线粒体内膜的荧光标志物。NAO一致认为能结合带负电荷的磷脂。NAO之所以能细胞内累积,与其同线粒体膜蛋白和/或脂质比如心磷脂的特定作用相关,基本上不依赖于线粒体膜电位。凋亡方面,线粒体膜电位的存在能用罗丹明123标记,而线粒体结构和完整性可用NAO来评估。NAO可通过流式检测来分析线粒体,也能用来描述多药耐药特征,以及测定凋亡过程中线粒体质量的变化。

METHOD:1 × 105 FL5.12-AM cells were resuspended in 0.5 ml of complete medium containing 50 nM mitochondrial fluorescent dye acridine orange 10-nonyl bromide (NAO). After incubating of 30 min at 37 °C in dark, cells were immediately transferred to a tube on ice for analysis by flow cytometry. [Reference: Fan Y et al. METHOD:Cells were incubated with or without H33342 or H33258 in 0.5 ml of 109cell/L HL-60 suspension in 12×75 mm plastic tubes at 37°C with 5% CO2. After incubation of cultured cells at various times with different amounts of H33342 or H33258, the delta psi or mitochondrial mass were measured with DiOC6(3) ornonyl acridine orange (NAO), respectively, added to achievea final concentration of 20 nM and incubated for 5 min at room temperature.Stock solutions of H33342, H33258, 4 μM DiOC6(3), or 4 μM NAO were dissolved in dimethyl-sulfoxide(DMSO). Finally, 2.5 μL of propidium iodide (PI) (1.0 mg/L stock) was added for the last 30s of incubation to assess cell viability. List mode files were collected for 25,000 events with a Coulter XL flow cytometer. [Reference: Chen JC et al.
